T6 is a stress response protein belongs to sirtuin family. It is involved in different multiple molecular pathways that are linked to DNA repair, neurodegeneration, tumorigenesis, glycolysis, gluconeogenesis, cardiac hypertrophy, metabolism etc. The up-regulation and down-regulation of this histone deacetylase involve in different diseases such as Alzheimer’s disease, aging, cancer, inflammation, diabetes, and cardiac problems. Thus, it has emerged as an important therapeutic target for the mentioned diseases. This review aims to compile available information regarding SIRT6 and its effect on smooth muscle- and epithelial tissues for predicting future target therapeutic approach. This review information exhibited that increased SIRT6 activity helps to combat diabetic atherosclerotic plaque; heal wound; inhibit EMT; save from pulmonary, cardiac, biliary and renal cell injury; decrease metabolic disease; glioma cell proliferation whereas decreased SIRT6 activity may improve Parkinson’s disease. Therapeutic approach can be developed by looking these sorts of effects of SIRT6 in cellular and molecular level.en_US
