Ekramul; Department of PharmacyThe research study was conducted to determine the antitumor effect of the flower of Alangium salvifolium (crude extract and diethylether fractions) against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 10 mg kg-1 body weight intraperitoneally. Extract/fractions was administered for nine consecutive days. Twenty-four hours of last dose and 18 h of fasting, the mice were sacrificed and antitumor effect was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight and hematological parameters of EAC bearing host. Significant (p<0.001) increases of survival times 30±0.96 and 25±0.40 days for crude extract and diethylether fraction of the A. salvifolium (10 mg kg-1) treated tumor bearing mice, respectively were confirmed with respect to the control group (20±0.13 days). The extract/fraction also decreased the body weight of the EAC tumor bearing mice. Hematological studies reveal that the heamoglobin (Hb) content was decreased in EAC treated mice whereas restoration to near normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC count and increase in WBC counts in extract/fraction treated animals when compared to EAC treated animals. Hence the present investigation has designed to appraise antidiabetic and antioxidant principle of the methanolic extract of Marsilea quadrifolia (MEMQ). Methods: Hypoglycemic effect was evaluated in alloxan induced diabetic rat. The oral administration of plant extract at a dose of 300 mg/kg body weight was given to fasting glucose loaded rat with regard to normal control during 1 hr. study period and in alloxan induced (110 mg/kg body weight i.p.) diabetic rat in comparison with reference drug Metformin Hydrochloride (100 mg/kg) during 3 days study period. The antioxidant potential of MEMQ was checked by qualitative method and quantitatively through DPPH(1,1- diphenyl-2-picryl-hydrazyl) scavenging assay at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: Considerable drop in elevated blood glucose level was observed in the alloxan induced diabetic (p<0.05 & p<0.001) rat. At a dose of 300 mg/kg the extract showed glucose level reduction of 47.57% in alloxan induced rat while 44.38% was found for Metformin after 3 days. Antioxidant activity using DPPH was found to increase in a concentration dependent manner with an IC 50 value of 96.37 ± 3.62μg/ml higher than the standard one, IC 50 16.59±0.59μg/ml. Total phenolic content was found 165.75 ± 0.961 mg/g in GAE and the total antioxidant capacity was equivalents of ascorbic acid (224.90 ± 1.42 mg/g). Conclusions: In all ways the extract showed significant antidiabetic and antioxidative potency. Therefore, this study is designed to investigate its antioxidant, analgesic and CNS depressant effects. Materials and Methods: The antioxidant activity of methanolic extract of S. nodiflora (SN) was determined using Folin Ciocalteu reagent, phosphomolybdenum method, free radical scavenging activity by 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and the reducing power activity. Analgesic activity was done by acetic acid and formalin model and CNS depressant activity was measured by the hole cross and open field method. Results: The total phenols and total antioxidant capacity of SN was found to be 37.38 < 2.01 mg/g equivalent of gallic acid and 433.37 < 4.22 mg/gm equivalent of ascorbic acid. The percentage (%) scavenging of DPPH free radical was found to be concentration dependent and IC50 value was 125.89 < 2.23 µg/ml while the IC50 value of standard ascorbic acid was found to be 16.76 < 0.11 µg/ml. The reducing power of SN was found to be concentration dependent. Acetic acid and formalin models are peripherally acting analgesic methods. The oral administration of both doses (100 and 200 mg/kg b.wt.) of SN significantly (p<0.001) inhibited 40.06% and 61.83% writhing response induced by acetic acid whereas oral administration of the same doses of SN significantly (p<0.001) inhibited 56.96% and 62.60% itching response induced by formalin. The methanolic extract of SN, at the dose of 250 mg/kg and 500 mg/kg b.wt. produced significant (P<0.001) decrease of locomotion. Protective effect of the ethyl acetate fraction (EAF) on H2O2-induced DNA damage in HepG2 cells were also evaluated, and the phytochemical profile of EAF was analyzed using tandem mass spectrometry. EAF derived from the 80% methanol extract of the leaf contained a remarkable amount of polyphenol and showed high levels of DPPH and alkyl radical scavenging activity, promoted cell viability, and protected against H2O2-induced DNA damage in HepG2 cells. Phytochemical analysis revealed that the major components in the EAF included quercetin monoglycoside, phloridizin, quercetin 3-diglycoside, quercetin-3-glucuronide, 2,6-dihydroxy-3,5-dimethyl-4-glucopyranosylbenzophenone, phenolic acid, guaijaverin, and naringin. The present study suggests possible synergistic or competitive antioxidant action of the major compounds of cattley guava leaf on H2O2-induced DNA damage in HepG2 cells. These results indicate that the ethyl acetate fraction of the guava leaf could be used as a potential source of natural antioxidants, and these findings will facilitate the utilization of guava leaf as a source of functional food.listelement.badge.dso-type Item , Antioxidant and antitumor activities of Polyalthia simiarum (Buch.-Ham. ex Hook. Antioxidant potential of the EA extract was evaluated in vitro by DPPH (1,l-diphenyl-2-picrylhydrazyl) scavenging assay method. The EA extract showed free radical scavenging activity with IC50 value of 24.08 ± 0.30 fig/mL. The antitumor activity of the extract was determined against Ehrlich ascites carcinoma (EAC) in mice at 25 mg/kg and 5Í) mg/kg body weight intraperitoneal. Significant (p<0.001) increase of survival time by 25 ± 0.57 and 27 ± 0.40 days by the EA extract treated tumor bearing mice was confirmed with respect to the control group (22 ± 0.12 days). Hematological studies revealed that the hemoglobin (Hb) content was decreased in EAC treated mice whereas restoration to close to normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC and increase in WBC counts in extract treated animals when compared to EAC affected animals. Therefore, the EA extract of P. simiarum was capable to exhibit moderate antioxidant and antitumor activities. Glucose control plays an important role in the pro-oxidant/antioxidant balance. Some antidiabetic agents may by themselves have antioxidant properties independently of their role on glucose control. Objective: The present investigation draws a comparison of the protective antioxidant activity, total phenol content and the antihyperglycemic activity of the methanolic extract of Cajanus cajan root (MCC) and Tamarindus indica seeds (MTI). Materials and Methods: Antidiabetic potentials of the plant extracts were evaluated in alloxan-induced diabetic Swiss albino mice. The plant extracts at the doses of 200 and 400 mg/kg body weight was orally administered for glucose tolerance test during 1-hour study and hypoglycemic effect during 5-day study period in comparison with reference drug Metformin HCl (50 mg/kg). In vitro antioxidant potential of MCC and MTI was investigated by using 1, 1- diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: There was a significant decrease in fasting serum glucose level (P < 0.001), reduction in blood glucose level (P < 0.001) in 5-days study, observed in the alloxan-induced diabetic mice. The reduction efficacy of blood glucose level of both the extracts is proportional to their dose but MCC is more potent than MTI. Antioxidant study and quantification of phenolic compound of both the extracts revealed that they have high antioxidant capacity. Conclusion: These studies showed that MCC and MTI have both hypoglycemic and antioxidant potential but MCC is more potent than MTI. The present study suggests that both MCC and MTI could be used in managing oxidative stress.
