    Hypoglycemic and in vitro antioxidant effects of methanolic extract of Marsilea quadrifolia Plant

    Introduction: Oxidative stress induced by alloxan has been shown to damage pancreatic beta cell and produce hyperglycemia in rats. Hence the present investigation has designed to appraise antidiabetic and antioxidant principle of the methanolic extract of Marsilea quadrifolia (MEMQ). Methods: Hypoglycemic effect was evaluated in alloxan induced diabetic rat. The oral administration of plant extract at a dose of 300 mg/kg body weight was given to fasting glucose loaded rat with regard to normal control during 1 hr. study period and in alloxan induced (110 mg/kg body weight i.p.) diabetic rat in comparison with reference drug Metformin Hydrochloride (100 mg/kg) during 3 days study period. The antioxidant potential of MEMQ was checked by qualitative method and quantitatively through DPPH(1,1- diphenyl-2-picryl-hydrazyl) scavenging assay at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: Considerable drop in elevated blood glucose level was observed in the alloxan induced diabetic (p<0.05 & p<0.001) rat. At a dose of 300 mg/kg the extract showed glucose level reduction of 47.57% in alloxan induced rat while 44.38% was found for Metformin after 3 days. Antioxidant activity using DPPH was found to increase in a concentration dependent manner with an IC 50 value of 96.37 ± 3.62μg/ml higher than the standard one, IC 50 16.59±0.59μg/ml. Total phenolic content was found 165.75 ± 0.961 mg/g in GAE and the total antioxidant capacity was equivalents of ascorbic acid (224.90 ± 1.42 mg/g). Conclusions: In all ways the extract showed significant antidiabetic and antioxidative potency. The present investigation suggests that MEMQ may be a potential source of natural antioxidant with good hypoglycemic activity.

    Anti-inflammatory and anti-diarrheal effects of methanolic extracts of seeds and peel of Nephelium longan fruits in rats

    Herbal medicines are playing a vital role in the remedy of numerous diseases. Preliminary phytochemical screening of the methanolic extracts of seeds (MNLS) and peel (MNLP) of Nephelium longan revealed the presence of alkaloids, tannins and flavonoids. For antidairrheal screening, we have followed Castoroil- induced method whereas to check anti-inflammatory property, we used carrageenan-iduced rat paw edema in Long-Evan rats. Two doses (250 and 500 mg/Kg) of the extracts and one dose of indomethacin as reference was used. Both extracts significantly (p < 0.05) reduced the formation of oedema induced by carrageenan in a dose-depending manner. MNLP and MNLS also exhibited anti-diarrheal action in dosedepending manner and all the results were found to be significant (p < 0.05) in comparison to loperamide. The outcomes indicate the potent anti-inflammatory and anti-diarrhoeal effects of N longan extracts on living models which are comparable with those of standard drugs such as indomethacin and loperamide respectively and support their conventional uses as medicine.

    Antidiabetic and antidiarrheal effects of the methanolic extract of Phyllanthus reticulatus leaves in mice

    Objective To assess the anti-diabetic and antidiarrheal activity of methanolic extract of Phyllanthus reticulates (P. reticulates) leaves in an animal model. Methods Phytochemical screening of methanolic extract of P. reticulatus leaves has been performed. Antidiabetic activity have been done by OGTT, normoglycemic hyperglycemia and alloxan induced diabetic mice. Plant extracts (150 mg/kg and 300 mg/kg, b.w.) were administered orally in fasting glucose loaded mice with regard to normal control and in alloxan induced (110 mg/kg body weight i.p.) diabetic mice in comparison with reference drug Metformin hydrochloride (100 mg/kg) during 7 day test period. Antidiarrheal test was conducted by castor oil and magnesium sulfate. Results Findings confirmed that the continuous post-treatment for 7 days with both extracts showed significant (P < 0.05) hypoglycemic activity in OGTT, normoglycemic and alloxan induced mouse models. Castor oil and Magnesium sulfate induced diarrheal test of the extract (200 and 400 mg/kg) has given significant effect in compairing to control diarrheal group. Conclusion Methanolic extract of P. reticulatus leaves have shown significant antidiabetic and antidiarrheal properties.

    Central nervous system depressant, analgesic and antidiarrheal effects of the seed extracts of Dimocarpus longan lour in rats

    Purpose: To assess the central nervous system (CNS) depressant, analgesic and antidiarrheal activities of the dried seed crude extracts of Dimocarpus longan Lour in rodents. Methods: Selected pharmacological effects of the ethanol (ENLS), petroleum ether (PELS), chloroform (CHLS) and ethyl acetate (EALS) extracts of D. longan fruit seeds were investigated. CNS depressant activity was evaluated by open field and hole cross tests; analgesic activity by acetic acid-induced writhing test and formalin-induced licking test; and anti-diarrheal activity was assessed in castor oil and magnesium-induced diarrhea rat model. The extracts were given orally in a rat model at doses of 200 and 300 mg/kg body weight. Normal saline served as control in all experiment. In CNS depressant test, diazepam (1 mg/kg) was used as reference drug while indomethacin (10 mg/kg) and loperamide(2 mg/kg) were used as standard drugs in analgesic and antidiarrheal tests, respectively. Results: In hole cross method, EALS showed the most effective depressant effect, viz, 1.17±0.17 for 200 mg/kg dose and 0.83±0.31 number of movements for 300 mg/kg dose after 120 min (p < 0.01), whereas in the open field test, all the extracts exhibited significant (p < 0.01) depressant effect in relation to positive control, diazepam. In acetic acid-induced pain test, PELS gave the lowest number of writhing (2.83±0.307) and the highest inhibition (88.45 %, 300 mg/kg dose) which was statistically significant. All the extracts also significantly (p < 0.01) suppressed licking activity in both phases of the formalin-induced licking test, in contrast to indomethacin. In the antidiarrheal tests, diarrheal suppression was highest at 300 mg/kg dose for all the extracts, compared with loperamide in both castor oil and magnesium sulphate induced diarrhea model. Conclusion: The extracts of Dimocarpus longan tested demonstrated significant CNS depressant, analgesic and antidiarrheal activities in a rodent model.
