In DPPH free radical scavenging assay, the extract exhibited potent antioxidant activity with a IC50 values of 2.43±1.06 μg/ml while in ascorbic acid, the value become 50 2.14±0.11 μg/ml. In thrombolytic activity using in vitro clot lysis assay method, the crude methanolic extract was found to have significant (p<0.001) thrombolytic activity at a dose of 10 mg/ml with a miximum effect of 20.23±1.56% while the standard streptokinase showed 46.13±3.87%. The extract was also investigated for its antibacterial and toxic potentiality using agar diffusion and Brine Shrimp lethality bioassay, respectively. The highest antibacterial effect was shown against Bacillus cereus (zone of inhibition 12±0.65 mm) followed by Escherichia coli (zone of inhibition 10±0.71 mm). In this bioassay the extract showed significant toxicity to Brine Shrimp nauplii with the LC50 value of 2.24±0.98 μg/ml. The different assay methods, including total antioxidant activity, scavenging free radical, authentic peroxynitrite and reducing power assessment were used to evaluate the antioxidant potential of the crude extract and its organic fractions. While antimicrobial activities were evaluated using agar disc diffusion method and measuring their MIC value. The EtOAc fraction, showed strong activity in all the model systems tested and in peroxynitrite model this fraction (IC50= 1.29±0.18 μg/ml) exerted four-fold stronger activity than standard penicillamine (IC50 value of 5.20±0.32 μg/ml). The reducing power of the extract was found to be concentration dependent. 50 The administration of the extract/fractions at a dose of 200 and 400 mg/kg body weight to the male Wistar rats increased the percentage inhibition of reduced glutathione, superoxide dismutase and catalase significantly. Whereas, lipid peroxidation level in hepatotoxic rats markedly decreased at a dose of 400 mg/kg body weight after 7 days. EtOAc fractions showed also the highest antibacterial effect against Staphylococcus aureus with zone of inhibition 30.00±0.25 mm. The Brine shrimp lethality bioassay was used to determine the toxicity of the extracts where dichloromethane (CH2 Cl2) fraction showed highest activity (LC50= 39.02±1.16 μg/ml). In addition, total phenolic and flavonoid content and total antioxidant capacity were also determined. Antioxidant potential of the extract/fractions was evaluated by DPPH (1,1diphenyl-2-picrylhydrazyl), NO (nitric oxide) and ONOO- (peroxynitrite) scavenging assay method. Ethyl acetate fractions (EtOAc) showed highest scavenging activity in all the methods with IC50 value of 12.98±0.08, 45.98±0.07 and 14.45±0.18 μg mL-1 for DPPH, NO and ONOO- assay method, respectively. In reducing power assay, EtOAc fraction also showed significant (p<0.001) activity. Further, the extract/fractions were studied for their analgesic (hot plate, tail immersion and acetic acid induced writhing test) and anti-inflammatory (carrageenan induced paw edema in rats) activities at a dose level of 200 and 400 mg kg-1 body weight. Among all the extract/fractions, EtOAc fraction showed a dose dependent and significant (p<0.005, p<0.05) analgesic activity in all the tested method. EtOAc fractions also reduced the paw edema considerably (86.80% inhibition after 3 h, p<0.005, p<0.05) in dose dependent manner compared to carrageenan induced rat. Antioxidant potential of the extract/fractions was evaluated by DPPH (1,ldiphenyl-2-picrylhydrazyl) and NO (nitric oxide) scavenging assay method. Ethyl acetate fractions (EtOAc) showed highest scavenging activity in all the methods with IC50 value of 12.32±0.16 and 5.38±0.07 pg mL -1 for DPPH and NO assay method, respectively. In reducing power assay, EtOAc fraction also showed significant (p<O.OOl) activity. Further, the extract/fractions were studied for their anti-inflammatory (carrageenan induced paw edema in rats) and antipyretic (Brewer's yeast induced pyrexia) activities at a dose level of 100, 200 and 400 mg kg-1 body weight for MeOH extract. Methanolic extract showed a dose dependent and significant (p<0.005, p<0.05) anti -inflammatory and antipyretic effect. Dichloromethane fraction (CH2CI2) and Ethyl acetate fractions exhibited similar activity using a dose of 200 mg kg-1 b.wt. in these models. The pharmacological activities of the (CH2CI2) fraction were lesser than the MeOH extract and other fractions. In addition, total phenolic and flavonoid content and total antioxidant capacity were also determined. Ekramul; Department of PharmacyThe research study was conducted to determine the antitumor effect of the flower of Alangium salvifolium (crude extract and diethylether fractions) against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 10 mg kg-1 body weight intraperitoneally. Extract/fractions was administered for nine consecutive days. Twenty-four hours of last dose and 18 h of fasting, the mice were sacrificed and antitumor effect was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight and hematological parameters of EAC bearing host. Significant (p<0.001) increases of survival times 30±0.96 and 25±0.40 days for crude extract and diethylether fraction of the A. salvifolium (10 mg kg-1) treated tumor bearing mice, respectively were confirmed with respect to the control group (20±0.13 days). The extract/fraction also decreased the body weight of the EAC tumor bearing mice. Hematological studies reveal that the heamoglobin (Hb) content was decreased in EAC treated mice whereas restoration to near normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC count and increase in WBC counts in extract/fraction treated animals when compared to EAC treated animals. Hence the present investigation has designed to appraise antidiabetic and antioxidant principle of the methanolic extract of Marsilea quadrifolia (MEMQ). Methods: Hypoglycemic effect was evaluated in alloxan induced diabetic rat. The oral administration of plant extract at a dose of 300 mg/kg body weight was given to fasting glucose loaded rat with regard to normal control during 1 hr. study period and in alloxan induced (110 mg/kg body weight i.p.) diabetic rat in comparison with reference drug Metformin Hydrochloride (100 mg/kg) during 3 days study period. The antioxidant potential of MEMQ was checked by qualitative method and quantitatively through DPPH(1,1- diphenyl-2-picryl-hydrazyl) scavenging assay at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: Considerable drop in elevated blood glucose level was observed in the alloxan induced diabetic (p<0.05 & p<0.001) rat. At a dose of 300 mg/kg the extract showed glucose level reduction of 47.57% in alloxan induced rat while 44.38% was found for Metformin after 3 days. Antioxidant activity using DPPH was found to increase in a concentration dependent manner with an IC 50 value of 96.37 ± 3.62μg/ml higher than the standard one, IC 50 16.59±0.59μg/ml. Total phenolic content was found 165.75 ± 0.961 mg/g in GAE and the total antioxidant capacity was equivalents of ascorbic acid (224.90 ± 1.42 mg/g). Conclusions: In all ways the extract showed significant antidiabetic and antioxidative potency. Methods: Two doses of the test compounds 25 and 50 mg/kg (p.o) for each were selected throughout the research work. The anti-inflammatory activity of the test compounds was determined by 'carragenan induced mice paw edema inhibition' method. The analgesic activity was determined by both, 'acetic acid induced writhing' and 'tail immersion' methods. All such data were compared with standard drugs at the dose of 10 mg/kg (p.o.). Results: Both ASC and BSC have showed positive effects as anti-inflammatory and analgesic agents. Anti-inflammatory and analgesic activities of the test compounds at 50 mg/kg (p.o.) were quite comparable to those of standard drugs at 10 mg/kg (p.o.). Therefore, this study is designed to investigate its antioxidant, analgesic and CNS depressant effects. Materials and Methods: The antioxidant activity of methanolic extract of S. nodiflora (SN) was determined using Folin Ciocalteu reagent, phosphomolybdenum method, free radical scavenging activity by 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and the reducing power activity. Analgesic activity was done by acetic acid and formalin model and CNS depressant activity was measured by the hole cross and open field method. Results: The total phenols and total antioxidant capacity of SN was found to be 37.38 < 2.01 mg/g equivalent of gallic acid and 433.37 < 4.22 mg/gm equivalent of ascorbic acid. The percentage (%) scavenging of DPPH free radical was found to be concentration dependent and IC50 value was 125.89 < 2.23 µg/ml while the IC50 value of standard ascorbic acid was found to be 16.76 < 0.11 µg/ml. The reducing power of SN was found to be concentration dependent. Acetic acid and formalin models are peripherally acting analgesic methods. The oral administration of both doses (100 and 200 mg/kg b.wt.) of SN significantly (p<0.001) inhibited 40.06% and 61.83% writhing response induced by acetic acid whereas oral administration of the same doses of SN significantly (p<0.001) inhibited 56.96% and 62.60% itching response induced by formalin. The methanolic extract of SN, at the dose of 250 mg/kg and 500 mg/kg b.wt. produced significant (P<0.001) decrease of locomotion. Antioxidant potential of the EA extract was evaluated in vitro by DPPH (1,l-diphenyl-2-picrylhydrazyl) scavenging assay method. The EA extract showed free radical scavenging activity with IC50 value of 24.08 ± 0.30 fig/mL. The antitumor activity of the extract was determined against Ehrlich ascites carcinoma (EAC) in mice at 25 mg/kg and 5Í) mg/kg body weight intraperitoneal. Significant (p<0.001) increase of survival time by 25 ± 0.57 and 27 ± 0.40 days by the EA extract treated tumor bearing mice was confirmed with respect to the control group (22 ± 0.12 days). Hematological studies revealed that the hemoglobin (Hb) content was decreased in EAC treated mice whereas restoration to close to normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC and increase in WBC counts in extract treated animals when compared to EAC affected animals. Therefore, the EA extract of P. simiarum was capable to exhibit moderate antioxidant and antitumor activities. This is the first report of antioxidant and antitumor potential of P. simiarum.
