Antioxidant potential of the extract/fractions was evaluated by DPPH (1,ldiphenyl-2-picrylhydrazyl) and NO (nitric oxide) scavenging assay method. Ethyl acetate fractions (EtOAc) showed highest scavenging activity in all the methods with IC50 value of 12.32±0.16 and 5.38±0.07 pg mL -1 for DPPH and NO assay method, respectively. In reducing power assay, EtOAc fraction also showed significant (p<O.OOl) activity. Further, the extract/fractions were studied for their anti-inflammatory (carrageenan induced paw edema in rats) and antipyretic (Brewer's yeast induced pyrexia) activities at a dose level of 100, 200 and 400 mg kg-1 body weight for MeOH extract. Methanolic extract showed a dose dependent and significant (p<0.005, p<0.05) anti -inflammatory and antipyretic effect. Dichloromethane fraction (CH2CI2) and Ethyl acetate fractions exhibited similar activity using a dose of 200 mg kg-1 b.wt. in these models. The pharmacological activities of the (CH2CI2) fraction were lesser than the MeOH extract and other fractions. In addition, total phenolic and flavonoid content and total antioxidant capacity were also determined. Hence, the present study was designed to investigate antidiarrheal, antioxidant and antibacterial potential of the methanolic extract of M.sapientum seed (MMSS). The extract was studied for antidiarrheal property using castor oil and magnesium sulfate induced diarrheal model and charcoal induced gastrointestinal motility test in mice. Total phenolic and flavonoids content, total antioxidant activity, scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, as well as nitric oxide (NO) and assessment of reducing power were used to evaluate antioxidant potential of MMSS. In addition, disc diffusion methods were used for antibacterial assay using various diarrheal induced bacterial strains. At the doses of 100 and 200 mg/kg body weight, the extract reduced the frequency and severity of diarrhea in test animals throughout the study period. At the same doses, the extracts significantly (p<0.001) delayed the intestinal transit of charcoal meal in test animals as compared to the control. In DPPH and NO scavenging method, MMSS showed good antioxidant potentiality in a dose dependent manner with the IC50 value of 12.32±0.33 μg/ml and 18.96±1.01 μg/ml, respectively with a significant (p<0.001) good reducing power. The extract also displayed strong antibacterial effect against when tested against Escherichia coli, Shigella dysenteriae, and Pseudomonas aeruginosa. The different assay methods, including total antioxidant activity, scavenging free radical, authentic peroxynitrite, nitric oxide and reducing power assessment were used to evaluate the antioxidant potential of the crude extract and its organic fractions. The ethylacetate (EtOAc) fraction, showed strong activity in all the model systems tested and in peroxynitrite model this fraction (IC50 value of 2.29±0.18 μg mL-1) exerted three-fold stronger activity than standard penicillamine (IC50 value of 6.20±0.32 μg mL-1). The reducing power of the extract was found to be concentration dependent. The administration of the extract/fractions at a dose of 250 and 500 mg kg-1 body weight to the male Wistar rats increased the percentage inhibition of reduced glutathione, superoxide dismutase and catalase significantly. Whereas, lipid peroxidation level in hepatotoxic rats markedly decreased at a dose of 500 mg kg-1 body weight after 7 days. The total phenol and flavonoid content were also measured in the crude extract along with its organic fractions. The Brine shrimp lethality bioassay was used to determine the toxicity of the extracts and Vincristin sulphate was used as positive control. The dichloromethane (CH2Cl2) fraction showed highest activity (LC50 value of 29.02±1.16 μg mL-1) and other showed activity in the order of: EtOAc fraction >n-BuOH fraction > MeOH extract > aqueous fraction. In DPPH free radical scavenging assay, the extract exhibited potent antioxidant activity with a IC50 values of 2.43±1.06 μg/ml while in ascorbic acid, the value become 50 2.14±0.11 μg/ml. In thrombolytic activity using in vitro clot lysis assay method, the crude methanolic extract was found to have significant (p<0.001) thrombolytic activity at a dose of 10 mg/ml with a miximum effect of 20.23±1.56% while the standard streptokinase showed 46.13±3.87%. The extract was also investigated for its antibacterial and toxic potentiality using agar diffusion and Brine Shrimp lethality bioassay, respectively. The highest antibacterial effect was shown against Bacillus cereus (zone of inhibition 12±0.65 mm) followed by Escherichia coli (zone of inhibition 10±0.71 mm). In this bioassay the extract showed significant toxicity to Brine Shrimp nauplii with the LC50 value of 2.24±0.98 μg/ml. Ekramul; Department of PharmacyIn the present study crude methanolic extract of Stephania japonica leaf was investigated for possible antioxidant, analgesic and toxic activity. The extract showed antioxidant activity in DPPH radical scavenging activity, nitric oxide scavenging activity and reducing power assays. In both DPPH radical and NO scavenging assay, the extract exhibited moderate antioxidant activity and the IC50 values in DPPH radical scavenging and NO scavenging assays were found to be 105.55±1.06 and 129.12±0.15 ug mL-1, respectively while the IC50 values of ascorbic acid were 12.30±0.11 and 18.64±0.22 ug mL-1, respectively. Reducing power activity of the extract increased in a dose dependent manner. Analgesic activity of the crude extract was evaluated using acetic acid-induced writhing model of pain in mice. The crude extract at 200 and 400 mg kg-1 b. wt. doses displayed significant (p<0.001) reduction in acetic acid induced writhing in mice with a maximum effect of 75.89% reduction at 400 mg kg-1 b.wt. which is comparable to the standard, diclofenac sodium (86.52%). The extract was also investigated for toxic potentiality using Brine Shrimp lethality bioassay. In this bioassay the extract showed significant toxicity to Brine Shrimp nauplii with the LC50 value of 25.19±0.98 ug mL-1. Antioxidant potential of the extract/fractions was evaluated by DPPH (1,1diphenyl-2-picrylhydrazyl), NO (nitric oxide) and ONOO- (peroxynitrite) scavenging assay method. Ethyl acetate fractions (EtOAc) showed highest scavenging activity in all the methods with IC50 value of 12.98±0.08, 45.98±0.07 and 14.45±0.18 μg mL-1 for DPPH, NO and ONOO- assay method, respectively. In reducing power assay, EtOAc fraction also showed significant (p<0.001) activity. Further, the extract/fractions were studied for their analgesic (hot plate, tail immersion and acetic acid induced writhing test) and anti-inflammatory (carrageenan induced paw edema in rats) activities at a dose level of 200 and 400 mg kg-1 body weight. Among all the extract/fractions, EtOAc fraction showed a dose dependent and significant (p<0.005, p<0.05) analgesic activity in all the tested method. EtOAc fractions also reduced the paw edema considerably (86.80% inhibition after 3 h, p<0.005, p<0.05) in dose dependent manner compared to carrageenan induced rat. Ekramul; Department of PharmacyThe research study was conducted to determine the antitumor effect of the flower of Alangium salvifolium (crude extract and diethylether fractions) against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 10 mg kg-1 body weight intraperitoneally. Extract/fractions was administered for nine consecutive days. Twenty-four hours of last dose and 18 h of fasting, the mice were sacrificed and antitumor effect was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight and hematological parameters of EAC bearing host. Significant (p<0.001) increases of survival times 30±0.96 and 25±0.40 days for crude extract and diethylether fraction of the A. salvifolium (10 mg kg-1) treated tumor bearing mice, respectively were confirmed with respect to the control group (20±0.13 days). The extract/fraction also decreased the body weight of the EAC tumor bearing mice. Hematological studies reveal that the heamoglobin (Hb) content was decreased in EAC treated mice whereas restoration to near normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC count and increase in WBC counts in extract/fraction treated animals when compared to EAC treated animals. Antioxidant potential of the OS extract was evaluated in vitro by DPPH (1, 1diphenyl-2-picrylhydrazyl) and NO (Nitric Oxide) scavenging assay and reducing power assay method. OS extract showed prominent scavenging activity in all the methods with IC50 value of 16.39±0.31 and 16.20±0.33 μg mL-1 for DPPH and NO assay method, respectively. In reducing power assay, OS extract also showed significant (p<0.001) activity. In addition, total phenolic and flavonoid content and total antioxidant capacity were also determined. The anti neoplastic effect of the leaves of OS against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 50 mg kg-1 body weight intraperit one ally. Significant (p<0.001) increases of survival times 33±1.81 days for crude extract of the (50 mg kg-1) treated tumor bearing mice were confirmed with respect to the control group (20±0.12 days). The extracts showed prominent DPPH free radical scavenging activity, inhibiting activity of authentic peroxynitrite (ONOO-) and inhibition of total reactive oxygen species (ROS) generation. In DPPH radical scavenging assay, the EtOAc fraction showed the highest activity with a IC50 value of 25.92±1.02 mg/ml whereas aqueous fractions had no activity at all within the tested concentration. Scavenging of the authentic ONOO- system, all extract/fractions showed good activity and among them, EtOAc fraction had the highest activity with a IC50 value of 15.79±0.18 mg/ml. EtOAc fraction also showed significant (p<0.001) inhibitory activity against the total ROS generation which was almost similar with that of the positive control Trolox (IC50 16.79±0.19mg/ml). Glucose control plays an important role in the pro-oxidant/antioxidant balance. Some antidiabetic agents may by themselves have antioxidant properties independently of their role on glucose control. Objective: The present investigation draws a comparison of the protective antioxidant activity, total phenol content and the antihyperglycemic activity of the methanolic extract of Cajanus cajan root (MCC) and Tamarindus indica seeds (MTI). Materials and Methods: Antidiabetic potentials of the plant extracts were evaluated in alloxan-induced diabetic Swiss albino mice. The plant extracts at the doses of 200 and 400 mg/kg body weight was orally administered for glucose tolerance test during 1-hour study and hypoglycemic effect during 5-day study period in comparison with reference drug Metformin HCl (50 mg/kg). In vitro antioxidant potential of MCC and MTI was investigated by using 1, 1- diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: There was a significant decrease in fasting serum glucose level (P < 0.001), reduction in blood glucose level (P < 0.001) in 5-days study, observed in the alloxan-induced diabetic mice. The reduction efficacy of blood glucose level of both the extracts is proportional to their dose but MCC is more potent than MTI. Antioxidant study and quantification of phenolic compound of both the extracts revealed that they have high antioxidant capacity. Conclusion: These studies showed that MCC and MTI have both hypoglycemic and antioxidant potential but MCC is more potent than MTI. The present study suggests that both MCC and MTI could be used in managing oxidative stress.
