dc.description.abstract | Shigellosis, a disease caused by Shigella spp. occur because of the presence of stx1 and stx2
genes. Shigellaspp. Sometimes escapes detection by the traditional culture methods. Bacterial
isolates were chosen on the basis of their structural, morphological and biochemical character.
To get the most efficient result along with traditional method PCR technique was used.
Antibiotic sensitivity testing was done to see the resistance of the isolates against Amoxicillin,
Ampicillin, Tetracyclin, Chloramphenicol, Norfloxacinantibiotic.To detect stx1, Ka1F and Ka1R
primers were used which yielded 348 bp fragment of stx1 gene. For stx2 gene Ka2F and Ka2R
primers were used to get 584 bp fragment of stx2 gene. The thermal profile for 35 cycles were
set using the following parameters 94°C for 10 minutes,94°C for 1 minute,55°C for 1
minute,72°C for 1 minute and 72°C for 7 minutes. This study was conducted to see the presence
of Shigella spp. on the environment. Also, to find out if the environmental Shigella spp. has stx1
and stx2 gene required to cause disease. | en_US |
